SPEAKER: Multiplex PCR is the concurrent detection of several DNA targets within a single well using different primers for each target. Multiplexing enables more sensitive detection of many targets from a small sample, saves money, and increases sample throughput. There are multiple types of multiplex PCR, including quantitative PCR or qPCR, and droplet digital PCR or ddPCR. DdPCR is a method based on water-oil emulsion droplet technology, which partitions nucleic acid samples into thousands of nano liter sized droplets.
SPEAKER: PCR amplification is then carried out within each droplet. Following PCR, each droplet is read to eventually determine target sequence concentration in the original sample. Since its introduction in 2011, ddPCR has been rapidly adopted by scientists to overcome the limitations of the traditional, real-time method, qPCR, by providing high performance with a simple and flexible workflow.
SPEAKER: One important limitation of qPCR is its limited accuracy in quantification of multiple targets in the same sample. ddPCR provides the unique ability to precisely measure three, five, or even 12 targets, using a multichannel system. ddPCR tools, such as Bio-Rad's qx600, have multiplexing capabilities, which provide more information from rare and precious starting materials, such as liquid biopsies, rare environmental samples, or compromised DNA without compromising the accuracy, sensitivity or precision of the ddPCR.
