SPEAKER 1: Cell proliferation is the rapid expansion of a cell population due to growth and division. Accurate measurement of proliferation is crucial in the assessment of cell health. It can be used to quantify responses to toxic insults, or as a prognostic and diagnostic tool in cancer. It's important in aging, drug resistance, drug discovery, immunology, and environment contamination research. Metabolic activity, cell proliferation markers, ATP concentration, and DNA synthesis are the main methods used for measuring cell proliferation.
SPEAKER 1: Measuring DNA synthesis is often the preferred method among researchers, as it offers a high level of precision for detecting changes in cell proliferation in vivo and in vitro. A popular technique for measuring DNA synthesis is the incorporation of BrdU, a synthetic thymidine analog into newly synthesized DNA in replication, which is then detected using anti-BrdU antibodies. For successful staining, it's important to include a DNA denaturing step to allow the antibody access to incorporated BrdU.
SPEAKER 1: Acids such as hydrochloric acid are used for this purpose. BrdU incorporation into cells and tissue can be visualized using various immunoassays, including immunohistochemistry, immunocytochemistry, and flow cytometry. This technique is highly translational and could be used for a number of applications, including drug screening and the analysis of anti-cancer medications, toxic substances, and other pharmaceuticals.
